Objectives This study aims to establish a protocol for a systematic review to assess the safety and effectiveness of manual therapy in treating carpal tunnel syndrome. Methods We will conduct a comprehensive search for relevant randomized controlled trials across seven databases, including MEDLINE/PubMed, Embase, the Cochrane Central Register of Controlled Trials, the China National Knowledge Infrastructure, Korean studies Information Service System, Oriental Medicine Advanced Searching Integrated System and Research Information Service System. Studies involving patients diagnosed with carpal tunnel syndrome and treated with manual therapy will be included. Comparators will include any other treatments, excluding manual therapy. Only randomized controlled trials evaluating manual therapy for carpal tunnel syndrome will be considered. Results Two independent researchers will screen studies based on inclusion criteria and extract the necessary data. Risk of bias will be assessed using the Cochrane risk of bias 2.0 tool. A meta-analysis will be performed if two or more studies measure the same outcome; otherwise, a qualitative analysis will be conducted. Subgroup analyses will be based on the type of manual therapy and the certainty of the evidence will be evaluated using the Grading of Recommendations, Assessment, Development, and Evaluation approach. Conclusions This systematic review will evaluate the effects of manual therapy on carpal tunnel syndrome. By reviewing the effectiveness of various manual therapy techniques, the study will aim to refine their clinical application and provide a foundation for future research.
